Combination Therapy of Cabozantinib, Nivolumab, and Ipilimumab in Advanced Renal-Cell Carcinoma Patients

Cabozantinib, nivolumab, and ipilimumab combination therapy have demonstrated significant progression-free survival benefits in previously untreated, advanced renal-cell carcinoma patients with intermediate or poor prognostic risk.

 

A phase 3 double-blind trial that enrolled 855 patients with advanced clear-cell renal-cell carcinoma who had not received prior treatment and had intermediate or poor prognostic risk randomized them into two groups: the experimental group (40 mg of cabozantinib along with nivolumab and ipilimumab daily), and the control group (matched placebo with nivolumab and ipilimumab).

 

The experimental group exhibited a higher progression-free survival probability at 12 months and a higher response rate compared to the control group. However, grade 3 or 4 adverse events were more common in the experimental group than in the control group.

 

Hence, the combination of Cabozantinib, nivolumab, and ipilimumab can potentially treat advanced renal-cell carcinoma patients who have not received treatment before and have intermediate or poor prognostic risk.
